We derive a method-independent universal precision bound for estimating the orientations of fluorescent molecules in the presence of Poisson shot noise using quantum detection and estimation theory. Combined with classical Fisher information and numerical analysis, we obtain a fundamental limit on the accuracy of measuring the ""wobble"", or rotational diffusion, of fluorescent molecules.
